The proper blocking/washing of nitrocellulose membranes to be used in rapid tests is often erroneously disregarded as a critical preparative step. For most systems, membrane blocking and washing improves sensitivity, specificity, and lot to lot consistency by (1) washing away potentially interfering production contaminates and unbound capture reagents, (2) preparing the membrane for optimal flow and (3) blocking sites on the nitrocellulose that could lead to unwanted background color "noice". Calypte offers a variety of well-tested membrane blockers including one with more “benign” surfactants for delicate systems and one that is does not contain proteins.
