Novel non-ionic surfactant-free diluent effectively blocks/washes nitrocellulose membrane used in rapid tests. Formulated with detergents better compatible with delicate systems normally affected deleteriously by typical nonionic detergents such as Tween® 20 (e.g. hydrophobic antigen systems). Improves sensitivity and specificity of most rapid test colloidal gold based systems. |

Use to block/wash nitrocellulose membrane after application of capture ligand. Appropriate for lateral flow, dot blot, and flow-through applications that are sensitive to typical nonionic detergents. |

Block nitrocellulose membrane only after applying the capture ligand and after it is completely dried (e.g. 4 hours at 40° C). Dilute Membrane Block MB-SF 1:10 in DI H2O to prepare a working 1X solution. Pour 1X solution into appropriate sized tray to a level of 1 cm and place 3-5 membrane vertically in the fluid to allow buffer to wick up the membrane in the direction of eventual sample flow. Do not allow membranes to touch one another during blocking. Remove membrane from buffer and dab the bottom edge to a paper towel to remove excess fluid. Incubate blocked membranes at 40° C for at least 4 hours (or overnight) to insure complete drying. Store blocked and dried membranes desiccated at ambient temperatures. |
