Block nitrocellulose membrane only after the capture reagent and after it is completely dried (e.g. 4 hours at 40° C ). Dilute Membrane Block MB-PF 1:10 in DI H2O to prepare a working 1 X solution. Pour 1X solution into appropriate sized tray to a level of 1 cm and place 3-5 membrane vertically in the fluid to allow buffer to wick up the membrane in the direction of eventual sample flow. Do not allow membranes to touch one another during blocking. Remove membrane from buffer and dab the bottom edge to a paper towel to remove excess fluid. Incubate blocked membranes at 40°C for at least 4 hours (or overnight) to insure complete drying. Store blocked and dried membranes desiccated at ambient temperatures. |
